When selecting a pack den leadership position for a user, I would think you would want to select the specific den leadership title (i.e. Wolf Den Leader) but I noticed there is a general “Den Leader”. What’s the difference and when would I select one over the other?

I should add that the official BSA position is “Den Leader”. The den they are a leader of is set by putting that person in the den. Just go to Manage -> Dens and drag them into the Wolf Den (or Bear, Webelos, etc).
We probably won’t be adding “Wolf Den Leader” as a position for this reason, but if you had other special positions we would consider them, like “Hall Monitor” or something like that.
